Scientific Background

The blend thesis behind KLOW is that its four peptides address complementary, largely non-overlapping nodes of tissue-repair biology, so a single vial covers angiogenesis, matrix remodeling and inflammation together in preclinical models. The component science is dose-independent, but the per-mL concentration after reconstitution depends on the exact vial loading.

• GHK-Cu delivers the matrix and antioxidant axis: at 1 to 10 nM it modulates collagen and glycosaminoglycan turnover and the MMP-TIMP system, induces bFGF and VEGF, and scavenges ROS (about 50% reduction of t-BHP-induced ROS in Caco-2 at 10 microM).
• TB-500, the N-acetylated active fragment Ac-LKKTETQ of thymosin beta-4, targets monomeric G-actin to buffer cytoskeletal dynamics, cell migration and angiogenesis.
• BPC-157 acts pro-angiogenically via VEGFR2-Akt-eNOS signaling, increasing vessel density and blood-flow recovery in ischemic rat hindlimb models (Hsieh 2017).
• KPV is taken up via the PepT1 transporter and inhibits NF-kB and MAPK signaling, lowering pro-inflammatory cytokines in murine colitis models (Dalmasso 2008).

Composition / Biochemical Properties

KLOW combines four structurally and mechanistically distinct peptides in a 25 : 10 : 10 : 10 ratio. Because it is a fixed blend, the four components cannot be dosed separately, and pharmacokinetics are component-specific rather than blend-wide.

• Composition: GHK-Cu (Gly-His-Lys with copper(II)) 25 mg, TB-500 fragment (Ac-LKKTETQ) 10 mg, BPC-157 (15-aa Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val) 10 mg, KPV (Lys-Pro-Val) 10 mg.
• Total mass: 55 mg per vial; GHK-Cu is the mass-dominant and least-stable component (rate-limiting for blend stability, optimal pH about 5.0 to 6.5).
• Half-life: only thymosin beta-4, the parent of TB-500, has hard human values (terminal t1/2 about 0.5 to 2.08 h; Wang 2021); GHK-Cu, BPC-157 and KPV have no validated human PK.
